Research Proven Health Benefits of Agaricus blazei Murill (ABM) Extract

1. Protect against infection
Agaricus blazei Murill (ABM) is a biologically active medicinal mushroom that has been used in traditional medicine and as a health food for prevention against infection and a range of diseases. ABM is rich in immunomodulating polysaccharides and beta-glucans, it helps to stimulate the immune cells, strengthen the immune system to help ward off pathogens.
In this animal test, Agaricus blazei Murill (ABM) extract or control was orally administered to some mice prior to infecting them with moderately virulent Streptococcus pneumoniae. Results showed that ABM extract protected the mice against S. pneumoniae, as shown by reduced bacteria in their blood as well as increased survival rate of the mice. This immune protection was suggested to be due to the involvement of ABM in native immune system. These findings suggest that ABM extract may be useful as a natural remedy to protect against bacterial and possibly other infections in humans.
Western equine encephalitis (WEE) is one of many mosquito-transmitted viral infections that can cause cytopathic effects, which is structural changes to host cells, leading to cell death. Some infected people, especially infants and elderly persons may develop brain inflammation. Among this, around 5 – 15% of brain inflammation cases are fatal, and about 50% of surviving infants would have permanent brain damage. There is no treatment for WEE other than supportive care.
In this in vitro study, the antiviral activities of Agaricus blazei Murill (ABM) extract on cells were investigated. The cells were cultured for 2 weeks after virus inoculation. ABM extract strongly suppressed the cytopathic effects induced by WEE virus, implying the beneficial effect of ABM extract on this viral infection.
2. Protect against allergy
Agaricus blazei have been shown to protect against allergy and asthma in animal models by modulating the T helper cell 1 (Th1) – T helper cell 2 (Th2) balance in the immune system. This medicinal mushroom upregulating Th1 response and downregulating Th2 response. During allergic reaction, the immune system overacts to an allergen by producing antibodies known as Immunoglobulin E (IgE), which is induced by a Th2 response, leading to symptoms in the nose, lungs, throat or on the skin.
In this animal test, mice were orally administered a mushroom extract, which consists mainly (82%) Agaricus blazei Murill (ABM) or control a day before or 19 days after the mice were induced allergen by being injected subcutaneously with model allergen ovalbumin (OVA). On day 26, their anti-OVA IgE (Th2 response) and IgG2a (Th1 response) antibodies were examined in the blood serum. Results showed that the mushroom extract given either before or after OVA immunization reduced the levels of anti-OVA IgE, but not IgG2a in the mice. This suggests that ABM extract may protect against allergy development and could potentially be used as a natural therapeutical substance against established allergy.
3. Provide antioxidant protection
Many mushrooms, including Agaricus blazei Murill (ABM) have been reported to provide antioxidant protection, which enables them to neutralise free radicals. The generation of free radicals can damage the cells, which appear to be a major contributor to ageing and degenerative diseases. Mushroom antioxidant components are found in the fruit bodies and mycelium, these include polysaccharides, phenolics, carotenoids and others.
In this in vitro study, the antioxidant properties of Agaricus blazei Murill (ABM) extract were evaluated. ABM extract provides antioxidant protection via three mechanisms. First, it inhibited enzymatic oxidative process, with 100% inhibition of horseradish peroxidase (HRP) and myeloperoxidase (MPO). Second, it inhibited cellular oxidative stress, with 80% inhibition of the oxidative burst of a type of white blood cells, known as polymorphonuclear neutrophils (PMNs). Third, it had direction action over reactive species, with 62% and 87% suppression of HOCl and superoxide anion radical, respectively. These findings suggest that ABM extract possesses significant antioxidant activity, indicating its potential application for nutraceutical and medicinal purposes.
5. Protect against paracetamol-induced brain and liver injury
Paracetamol is an over-the-counter medicine that is commonly used to relieve mild or moderate pain, such as headache, toothache or sprains and also reduce fever caused by colds and influenza. Overdose of paracetamol is toxic to the liver, it can lead to serious liver damage and possibly death. In addition to liver damage, paracetamol overdose is also toxic to the brain. It can induce morphological aberrations, implying the possibilities of higher brain impairments.
In this animal test, one group of mice was orally administered a single dose of paracetamol at 2 g/kg body weight, while another group was treated with the Agaricus blazei Murill (ABM) extract at 200 mg/kg body weight daily, for 21 days and paracetamol was given as a single dose of 2 g/kg body weight.
Results showed that the ABM pretreatment largely prevented the increase in the levels of liver enzymes in the blood plasma: aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH). Elevated AST, ALT, ALP and LDH could indicate liver damage or liver problems. ABM also normalised the TBARS levels in both the liver and brain tissues. TBARS (thiobarbituric acid reactive substances) are formed as a byproduct of lipid peroxidation and it is a widely used marker of oxidative stress. These findings indicate that ABM extract is able to provide a reasonable degree of protection against paracetamol-induced liver and brain injury.
6. Complement treatment therapy for chronic diseases
In this clinical trial, 100 patients with gynecologic diseaseswere treated either with carboplatin (300 mg/m2) plus VP16 (etoposide, 100 mg/m2) or with carboplatin (300 mg/m2) plus taxol (175 mg/m2) at 0, 3 and 6 weeks. ABM extract was orally administered every day. Compared to placebo group, ABM-treated group showed significantly higher natural killer (NK) cell activity. NK cells are immune cells which are capable of killing mutatedcells, detect and control early signs of diseases. In addition, ABM extract improved side effects of therapy, such as poor appetite, hair loss, emotional distress and general weakness. These findings suggest that ABM extract might be beneficial for patients with gynecologic disease who are undergoing treatment.
7. Normalise liver function in patients with chronic liver disease
In this 1-year open-label pilot study, patients with chronic liver disease for more than 3 years and not taking lamivudine, alpha-interferon or other drugs for chronic liver disease were orally administered 1500 mg Agarius blazei Murill (ABM) extract daily for 12 months. The level of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) was measured. High levels of AST and/or ALT may be a sign of liver damage or disease. At the end of the study, ABM extract was shown to result in a decrease of mean level of AST and ALT, indicating the potential benefit of ABM extract in normalising liver function of patients with chronic liver disease
8. Manage blood glucose levels
In this randomised, double-blind, placebo-controlled clinical trial, 72 type 2 diabetes patients with extremely high blood glucose levels who having been taking gliclazide and metformin for more than 6 months were orally administered either 1500 mg Agaricus blazei Murill (ABM) extract or placebo daily for 12 weeks. At the end of the study, patients received ABM extract showed significantly lower HOMA-IR index, which is an insulin resistance score. High HOMA-IR values indicate low insulin sensitivity (insulin resistance), while low HOMA-IR values indicate high insulin sensitivity.
Adiponectin is a protein hormone and adipokine that is involved in regulating glucose levels. In human, adiponectin levels in blood plasma are significantly lower in insulin-resistant states. In this study, oral administration of ABM extract for 12 weeks resulted in 20% increase in plasma adiponectin concentration, while those taking placebo showed 12% decrease. These findings suggest that ABM extract in combination with metformin and gliclazide reduces insulin resistance among subjects with high, unhealthy blood glucose levels.

Different mushrooms contain different types of polysaccharides with unique healing properties. The higher the polysaccharides content, the better the quality.
