Increase macular pigment optical density
In this clinical trial, healthy subjects aged 26 to 57 years were orally administered a supplement containing 20 mg/day of lutein, 4 mg/day of zeaxanthin and other antioxidants (vitamin C, vitamin E, zinc, copper) for 16 weeks and their macular pigment optical density (MPOD) levels and skin carotenoid levels were measured. By 8 weeks of intake, the total volume of MPOD significantly increased and continued to increase until week 16 (p <0.0001). Besides, the skin carotenoid levels significantly increased by week 4 and continued to increase until week 16 (p < 0.0001). These findings suggest the efficacy of lutein and zeaxanthin supplementation in improving MPOD and skin carotenoid levels.

Mitigate adverse effects associated with prolonged use of digital device
Study 1: In this clinical trial, healthy young adults with at least 6 hour of daily near-field screen time exposure were orally administered 24 mg macular carotenoids (MC) (lutein, zeaxanthin and meso-zeaxanthin) or placebo daily for 6 months and their effect on macular pigment (MP), visual performance, sleep quality and physical indicators of excessive screen time were determined. Compared to placebo, supplementation with macular carotenoids (MC) resulted in significant improvement in MPOD, overall sleep quality, headache frequency, eye strain, eye fatigue and all visual performance measures (p < 0.05). Besides, several undesirable physical outcomes associated with excessive screen time also improved with the supplementation. These findings suggest the beneficial effects of macular carotenoids (MC) in improving visual performance, sleep quality and adverse physical symptoms in those with high screen time exposure.

Study 2: In this clinical trial, healthy subjects (aged 24 - 51) were given 50 mg anthocyanins and resulted in improved dark adaptation. In another test, healthy subjects (aged 20 – 25) were given 50 mg anthocyanins and no decrease in the average refraction values, but a large decrease in placebo group was observed after a video display terminal (VDT) work.

Age-related macular degeneration (AMD) happens when a part of the retina, known as macula is damaged by aging, resulting in blurred central vision.
Study 1: In this clinical trial, patients with wet or dry age-related eye problem were orally administered 30 mg/day saffron or placebo for 6 months. After 3 months of saffron administration, there was significant increase in electroretinography (ERG) results (p = 0.027), which is a test that measures the electrical response of the light-sensitive cells in the eyes. At the follow-up, patients with wet age-related eye problem administered with saffron showed significant decrease in optical coherence tomography (OCT) results (p = 0.05), which is used as an early diagnosis of many serious eye conditions. These findings suggest saffron supplementation may help improve retinal function in patients with age-related eye problem.

Study 2: In this clinical trial, patients with early age-related eye problem were orally administered 20 mg/day saffron or placebo for 3 months, and then reverted to placebo or saffron for a further 3 months and focal electroretinograms (ERG) were recorded at baseline and after 3 months of supplementation. Focal ERG is a tool that measures the functional integrity of the central macula. Compared to placebo, there was significant increase in amplitude of focal ERG after saffron supplementation. Compared to baseline, focal ERG thresholds decreased after saffron supplementation, but not in placebo group. These findings suggest saffron supplementation helps improve retinal flicker sensitivity in early age-related eye problem.

Study 3: In this clinical trial, adults aged 50 years with mild/moderate age-related eye problem were orally administered 20 mg/day saffron or placebo for 3 months, followed by crossover for 3 months. Compared to placebo, saffron administration improved mean best-corrected visual acuity (BCVA) by 0.69 letters (p = 0.001) and reduced mean-pooled multifocal electroretinogram (mfERG) latency by 0.17 ms (p = 0.04). mfERG is an assessment of retinal function. These findings suggest saffron supplementation could help improve those with age-related eye problem. Study 1: In this clinical trial, young healthy subjects aged 18 to 25 years were orally administered 13 mg/day or 27 mg/day macular xanthophylls (MXans) (lutein, zeaxanthin and meso-zeaxanthin) or placebo for 6 months and their serum parameters and cognitiveperformance were determined at baseline and 6 months. Compared to placebo, administration of both 13 mg/day and 27 mg/day macular xanthophylls resulted in significant increase in serum macular xanthophylls, MPOD (macular pigment optical density), BDNF (brain-derived neurotrophic factor, which is essential for learning and memory) and antioxidant capacity (AOC) (p < 0.05). Besides, inflammatory cytokine IL-1β significantly decreased in both treatment groups, compared to placebo. For cognitive performance, the scores for composite memory, verbal memory, sustained attention, psychomotor speed, and processing speed improved significantly in both treatment groups, while remained unchanged in the placebo group. These findings suggest the efficacy of macular xanthophylls in improving MPOD and BDNF, which could help improve cognitive performance (Physiol Behav. 211: 112650 (2019)).
Study 2: In this clinical trial, patients with memory problem and cognitive issue were orally administered saffron for 12 months. Results showed that patients administered with saffron had improved Mini-Mental State Examination (MMSE) scores (p = 0.015), while the control group showed deterioration. MMSE is commonly used as a screening test for cognitive impairment. Besides, magnetic resonance imaging (MRI), electroencephalogram (EEG) and event-related potential (ERP) showed improvement in specific domains. These findings suggest the beneficial effect of saffron in the management of aMCImd.
Study 3: In this clinical trial, patients with mild to moderate memory concerns were orally administered 30 mg/day saffron or placebo for 16 weeks and their cognitive profiles were monitored. Compared to placebo, 16-weeek saffron administration resulted in better outcome on cognitive function. Patients administered with saffron showed lower ADAS-Cog (AD assessment scale-cognitive subscale) and CDR-SB (clinical dementia rating scale-sums of boxes) scores, indicating better cognitive performance. In term of observed adverse events, there were no significant differences in placebo and saffron groups. These findings suggest that saffron is safe and effective in the management of mild to moderate memory concerns.

Improve sleep quality
In this clinical trial, the subjects with mild to moderate sleep disorder associated with anxiety were orally administered saffron extract (15.5 mg/day) or placebo (maltodextrin) for 6 weeks. Their sleep quality was assessed using actigraphy, LSEQ and PSQI questionnaires. Results showed that 6-week saffron supplementation led to an increased time in bed assessed by actigraphy, improved ease of getting to sleep assessed by LSEQ questionnaire and improved sleep quality, sleep latency, sleep duration and global scores assessed by the PSQI questionnaire. A lower PSQI scores indicate better sleep quality. Meanwhile, those parameters were not modified in placebo group. These findings suggest saffron extract could be a safe and natural nutritional approach to improve sleep quality and duration.

Study 1: In this clinical trial, young healthy subjects were orally administered total 13 mg/day or 27 mg/day macular carotenoids (MC) (lutein, zeaxanthin and meso-zeaxanthin) or placebo for 12 months and their effects on blood cortisol, psychological stress ratings, behavioural measures of mood and symptoms of sub-optimal health were determined. Study 2: In this clinical trial, healthy adults aged 18 to 54 years old with subclinical feelings of low mood and anxiety and/or stress were orally administered 30 mg standardised saffron extract or placebo for 8 weeks. At the end of study, participants who administered with saffron extract reported reduced depression scores and improved social relationships. Besides, saffron supplementation resulted in significant increase in urinary crocetin levels, which is believed to reduce oxidative stress. Besides, saffron extract administration also attenuated typical stress-induced decreases in heart rate variability (HRV) during exposure to stressor. These findings suggest the efficacy of saffron extract in improving subclinical depressive symptoms in healthy individuals and may increase resilience against the development of stress-related psychiatric disorders.
