Study 1: Cancer patients
In this clinical trial, 34 advanced-stage cancer patients were orally administered with 1800 mg Ganoderma lucidum polysaccharide (GLPS), 3 times daily before meals for 12 weeks. Treatment for 12 weeks resulted in a significant increase in interleukin-2 (IL-2) and interferon-gamma (IFN-ɣ), which are cytokines responsible to induce and modulate an array of immune responses. In this clinical trial, male football players on a 28-day “living high-training low” (LHTL) trial were orally administered placebo or Ganoderma lucidum for 6 weeks and their CD4+/CD8+ ratio were determined. CD4+/CD8+ ratio is a reflection of immune health. A normal ratio is between 1 and 4. This ratio tends to decrease if the immune system’s defense against pathogens become weaker.
Due to simultaneous stimuli of physical activity and exposure to hypoxia (low oxygen), a significant decrease in CD4+/CD8+ ratio following the 28-day trial compared to the baseline was observed in placebo group. The volunteers were then asked to respond to SF-12 questionnaires for quality of life and score the levels of fatigue and VAS, before and 4, 8 and 12 weeks after the first dose. In addition, satisfaction of volunteers with 12-week G. lucidum extract was significantly higher than those taking placebo. In this clinical trial, 71 patients with type 2 diabetes were orally administered Ganoderma lucidum polysaccharides or placebo at 1800 mg 3 times daily for 12 weeks. Compared to baseline, G. lucidum polysaccharides significantly decreased haemoglobin A1c (HbA1C), which indicates the average blood glucose levels over the past 3 months. In addition, patients
treated with G. lucidum polysaccharides also showed a significant decrease in postprandial plasma glucose (PPG) levels, which is a measure of blood glucose level an hour or 2 after a meal. The lower HbA1C and PPG, the better the blood glucose control. These parameters did not change or slightly increased in patients administered with placebo. These findings suggest that G. lucidum polysaccharides are efficacious in lowering blood glucose levels.
6. Improve liver health
Reishi or Lingzhi (Ganoderma lucidum) is widely known in the treatment of chronic liver disease with little or no side effects, attributed to its antioxidant, anti-inflammatory and hepatoprotective properties. In this animal test, rats were induced to have liver fibrosis by gamma irradiation and CCl4, and oral co-administration of 110mg/kg Ganoderma lucidum. G. lucidum provided remarkably liver protection as seen by significant decrement in liver enzymes aspartate aminotransferase (AST) and alanine aminotransferase (ALT). An elevation of liver enzymes may be a sign of a liver problem. In addition, G. lucidum also decreased levels of malondialdehye (MDA), an indicator of liver damage, as well as increased antioxidant enzyme (GSH, SOD) activities. Microscopical examination also revealed suppression of G. lucidum on liver fibrosis. These findings support protective effects of G. lucidum against liver fibrosis.

The polysaccharides significantly (p < 0.01) remodeled gut microbiota compositions. Bacteria that are known for anti-obesity, SCFA-producer, polysaccharides/xylan degradation, and lactic acid production were significantly (p < 0.05) abundant in mice treated with polysaccharides.
